ROLE ’s Off-the-Shelf T cell project is focused on the development of an iPSC derived T-cell product. The project team is growing rapidly to enable delivery of a ground-breaking new therapy and ensure the company’s position as a leader in the allogeneic cell therapy field. Gene editing is an integral part of the process and the gene editing team is expanding. A Principal Scientist is required to join the gene editing team to help support the following duties.• Evaluation and optimisation of novel gene editing methodologies.• Contribute to pipeline development for the analysis of off target gene editing events.• Implement unbiased genome wide assays for the analysis of off target editing events.• Contribute to the bioinformatics strategy of the gene editing team.• Training junior members of the team in editing methodologies, associated assays and data analysis. • Gene editing in iPSC lines.• Molecular characterisation of edited iPSC lines.• Working extremely closely with colleagues in other R & D teams as part of a cross functional project team.Reporting to Group Leader/Associate Director of gene editing, the purpose of this role is to primarily conduct scientific experiments to support either the departmental objectives or company project objectives as necessary within dedicated scientific or project team.It is expected that a Principal Scientist may function as a specialist in a particular area or discipline and enable execution of experiments within a group, through designing, conducting and co-ordinating experiments, generating and interpreting data and contributing to the group’s and/or project’s scientific and experimental discussions. However, cross-discipline/functional thinking is also an important aspect of the Principal Scientist’s job. This role will require partnering with the Group Leader and other Principal Scientists to help achieve the strategic goals of the company and ensure the optimal flow of data, as required by function.The Principal Scientist may also hold responsibilities for tutoring, mentoring and supervising scientists. Working with the Team Lead you will be responsible for day to day experimental oversight of staff within the team.KEY RESPONSIBILITIES AND ACCOUNTIBILITIES:• Knowledge: Accountable for seeking, understanding and “distributing” knowledge and being at the fore-front of the gene editing field. Reading, attending conferences, etc. and communicating learnings to others• Scientific Expertise: Designing experiments for the group and/or project members and helping to problem solve scientific issues• Scientific integrity: Risk assessing department or project progress and experimental contingency planning, making recommendations to the group, project teams and key stakeholders (as appropriate). Ensuring a high standard of production, quality assessment and interpretation of data from the wider team.• Scientific Leadership: Act as the scientific champion within the gene editing field, enabling the company to achieve its objectives. Evolving appropriate protocol development and identifying the need for company policies. Driving innovation and new technology adoption with the function. Mentoring and tutoring scientists.• Supervision: Providing experimental supervision and support to the group/project members, including, in partnership with Group Leaders, input into objective setting. Providing detailed feedback to the Group Leaders, department heads etc. regarding the scientific progress on group members.• External: Act as a company representative at scientific conference actively seeking to present company data (talks and posters) in line with company publication strategy. Actively seeking to publish data where appropriate. Provision of technical support of external collaborations and responsibility for third party interactions.QUALIFICATIONS & E • Undergraduate degree in Life Sciences or related discipline.• Post graduate degree preferably including an MSc or PhD in a relevant scientific area.• Extensive Post-graduate experience in an academic or industrial setting.• Comprehensive understanding and practical experience of gene editing with different platforms and molecular characterisation of edited cells. Optimisation of gene targeting strategies.• Up to date knowledge, experience and data interpretation of unbiased genome wide assays for the discovery of off target editing events.• Interpretation of next generation sequencing data. • Gene editing in primary cells line, preferably iPSC.• Enthusiasm for technical challenges.• Demonstrable ability to design experiments and studies to address key scientific questions.• Extensive experience of all relevant laboratory procedures and protocols, with high competence at operating all applicable/specialist equipment and modifying procedures to suit the business needsDesirable • Experience of or exposure to driving projects forward (not the operational aspects associated with project management), including proven success in scientific project coordination from inception to conclusion.• Involvement in supervision of, or training support for scientists. • Gene editing with a focus on translation/development of cellular therapies.• Whole genome sequencing.• Editing with viral or non-viral supplied repair templates.SKILLS & COM • Molecular Biology skills – DNA/RNA purification, cloning, PCR, qPCR, DNA sequence analysis.• Gene editing and characterisation of off target events• Excellent communication skills and the ability to communicate to a variety of audiences including senior management, project teams etc.• Aptitude to design experiments and studies to address key scientific questions and be involved with scientific decisions of the group• High attention to detail• Proven scientific drive and sound scientific judgement, technical ability. • Autonomy, authority and confidence in scientific decision making.• Strong time management skills.• Ability to work effectively in a team.• Ability to lead teams, groups and/or projects (skills separate to those required by line management).• Skills involving influencing and management upwards (senior staff members), sideways (peers) and downwards (junior scientists).• Ability to work autonomously.• Aptitude to troubleshoot problems and create viable solutions in a constructive manner. • Strong problem solving abilities, good time management and attention to detail.• Strong team player and proactive communication with the project team.Desirable• Familiarity with flow cytometry or alternative single cell isolation approaches.• Background in T cell Biology/Immunology.• Stem cell culture.Abingdon, Oxfordshire, United Kingdom Full Time Allogeneic Experienced
